O'Hair was born in Pittsburgh, Pennsylvania, in 1918. Her parents were devout Catholics, but she rejected religion at a young age. She became an atheist after reading a book by Clarence Darrow.

In 1963, O'Hair founded American Atheists. The organization's mission is to promote atheism and to defend the separation of church and state. O'Hair was a vocal critic of organized religion, and she often made headlines with her controversial statements.

In 1969, O'Hair won a landmark Supreme Court case that prohibited the teaching of creationism in public schools. She also won a case that allowed atheists to hold public office. O'Hair's activism helped to raise the profile of atheism in America.

However, O'Hair's outspokenness also made her a target for threats and violence. In 1995, she was assassinated by three former members of American Atheists. Her murder remains unsolved.
